(1) During stocking time for the soda vending machine, I normally do not touch anything, just fill it up, it maybe takes 10min or so.
I am wondering what would be the best during stocking time, do I need to turn off vending machine? do I need to lower switch? or it doesn't matter?

(2) How coldness would be effective? during summer==> strong?, winter==> weak? spring/fall=> medium?

There's no need to turn off power to the machine when restocking. Simply do as you've been doing and you'll be just fine. I'd add that it would be good to thoroughly vacuum out the dust at the bottom of the machine monthly to keep it running as efficiently as possible. My machine eats about $25/month in electricity.

The recommended temp is 38 degrees. I've had mine at 42 degrees for more than a year without a single complaint. I wouldn't recommend adjusting the temp seasonally. When a customer wants a cold soda that's what they should get no matter the season.
